چکیده

Introduction Phosphorus-based peptidomimetics have been attractive target compounds during research and development of metalloproteinase inhibitors [1]. Their inhibiting activities are thought to be associated with mimicry of the metal-mediated transition state or tetrahedral-intermediate of the amide hydrolysis [2]. Because of the important roles of metal ions in the structures and functions of metalloproteinases, we envision that systematic investigation of the metal binding properties of phosphorus-based peptidomimetics should provide better understanding of their mechanism of action and assist the rational design of potent and specific inhibitors.

Design, synthesis, and metal binding of novel Pseudo- oligopeptides containing two phosphinic acid groups.

Phosphinic compounds have potential as amide-bond mimetics in the development of novel peptidomimetics, enzyme inhibitors, and metal-binding ligands. Novel pseudo-oligopeptides with two phosphinic acid groups embedded in the peptide backbone serving as amide-bond surrogates, Psi[P(O,OH)--CH(2)], were targeted. A series of linear and cyclic pseudo-oligopeptides with two phosphinic acid groups ar...

Enhanced Bioadsorption of Cadmium and Nickel by E. coli Displaying A Metal Binding Motif Using CS3 Fimbriae

Display of peptides on the surface of bacteria offers many new and exciting applications in biotechnology. Fimbriae is a good candidate for epitope display on the surface of bacteria. The potential of CS3 fimbriae of enterotoxigenic E. coli as a display system has been investigated. A novel cell surface display system with metal binding property was developed by using CS3 fimbriae. Short metal ...

Functional, metal-based crosslinkers for α-helix induction in short peptides.

Many protein-protein interactions that play a central role in cellular processes involve α-helical domains. Consequently, there has been great interest in developing strategies for stabilizing short peptides in α-helical conformations toward the inhibition and interrogation of protein-protein interactions. Here, we show that tridentate Hybrid Coordination Motifs (HCMs), which consist of a natur...

Peptide-bond modification for metal coordination: peptides containing two hydroxamate groups.

Peptide-bond modification via N-hydroxylation has been explored as a strategy for metal coordination to induce conformational rigidity and orient side chains for specific molecular recognition. N-Hydroxyamides were prepared by reacting N-benzyloxyamino acid esters or amides with Fmoc-AA-Cl/AgCN (Fmoc: 9-fluorenylmethoxycarbonyl; AA: amino acid) in toluene or Fmoc-AA/HATU/DIEA in DMF (HATU: O-(7...

Zn(II) and Hg(II) binding to a designed peptide that accommodates different coordination geometries.

Designed metal ion binding peptides offer a variety of applications in both basic science as model systems of more complex metalloproteins, and in biotechnology, e.g. in bioremediation of toxic metal ions, biomining or as artificial enzymes. In this work a peptide (HS: Ac-SCHGDQGSDCSI-NH2) has been specifically designed for binding of both Zn(II) and Hg(II), i.e. metal ions with different prefe...
